Two sample traces are produced on each sample filter. Options for analysis include:

  • Analyze one trace and archive the second trace
  • Analyze one trace for fungal spores and culture the second trace
  • Analyze both traces using the same method for increased sensitivity

A "key and slot" is included in the perimeter of the cassette, so that the cassette only fits together one way. This allows field personnel to remove one trace for microscopic analysis, re-assemble the cassette, and send the second trace to a laboratory for culturing or other analysis. If the small end cap is removed, the large end cap may be used to support the cassette upright.

  1. Cut through the label securing the two halves of the cassette
  2. Position the cassette so that the slotted inlet is facing up
  3. Remove the top portion of the cassette
  4. Place the filter, on the support pad, onto a clean surface
  5. Cut the filter in half between the two sample traces

Microscopic Analysis: MCE filter + Acetone vapor

  1. Place one sample trace (half-filter) on a glass slide
  2. Insert the half-filter and slide into an acetone vaporizer and clear the filter
  3. Place a drop of 3% KOH or similar liquid on the cleared filter
  4. Cover the sample with a cover slip
  5. Analyze the sample trace at a minimum magnification of 600X using light microscopy
 

 

Microscopic Analysis: MCE filter + Triacetin

  1. Place several drops of Triacetin on a glass slide
  2. Place one sample trace (half-filter) on the Triacetin
  3. Gently heat the filter using an alcohol lamp until clear (several seconds)
  4. Cover the sample with a cover slip
  5. Analyze the sample trace at a minimum magnification of 600X using light microscopy

  1. Culture one sample trace (half-filter) using standard dilution plating techniques
